Of course not. You are lost again when you turn your back on Christ, systematically stop trying, and either forget Him altogether or take Him for granted and "sin, that grace may abound."

Naturally any time you repent He will forgive you, and no, one sin does not constitute being lost, however grave it is in itself; but a tiny sin can lead to it if it starts you down a road away from Christ, by deliberately disobeying.
